目的:观察过量氟对牙胚组织中TGF-β1和AMTN表达的影响。方法:采用Trowell支架法体外分离和培养大鼠牙胚组织。将牙胚分别暴露于含0(对照组)、0.5、1.0、4.0 mmol/L氟化钠的培养基,染氟1、3、7 d后,采用RT-q PCR技术检测细胞内TGF-β1和AMTN相对表达水平的变化。结果:各染氟组牙胚中TGF-β1和AMTN的表达较对照组均明显降低(P<0.05),并呈剂量效应和时间效应;染氟组中TGF-β1和AMTN mRNA相对表达水平二者呈明显的正相关。结论:在釉质发育起始阶段,过量氟抑制牙胚中TGF-β1和AMTN的表达是氟致牙釉质矿化缺陷的重要病理机制。
Objective: To observe the effect of excessive fluoride on the expression of TGF-β1 and AMTN in tooth germ. Methods: Rat tooth germ tissues were isolated and cultured by Trowell method in vitro. The tooth germs were exposed to medium containing 0 (control group), 0.5, 1.0 and 4.0 mmol / L sodium fluoride, respectively. After being exposed to fluoride for 1, 3, and 7 d, RT-q PCR was used to detect the expression of TGF- And AMTN relative expression level changes. Results: The expression of TGF-β1 and AMTN in dental fluorosis group were significantly lower than those in control group (P <0.05), and the dose-effect and time-effect of TGF- Who showed a clear positive correlation. Conclusion: In the initial stage of enamel development, excessive fluoride inhibits the expression of TGF-β1 and AMTN in tooth germ, which is an important pathological mechanism of fluoride-induced enamel mineralization.
