目的:探讨蜣螂乙醇提取物抗大鼠前列腺增生的影响及其可能机制。方法:去势雄性大鼠皮下注射丙酸睾酮造成良性前列腺增生模型,将模型大鼠分为3组,分别灌胃蜣螂提取液、非那雄胺水溶液和同体积蒸馏水,正常对照组和模型组给予等体积蒸馏水,每天给药一次,连续给药4周,末次给药1 h后,处死大鼠,检测大鼠血清睾酮、雌二醇水平与前列腺指数,观察前列腺组织形态,采用免疫组化法检测表皮生长因子(EGF)、转化生长因子-β1(TGF-β1)在大鼠前列腺组织中的表达情况。结果:蜣螂20g生药/kg使造模大鼠血清睾酮水平增加、雌二醇水平降低;同时使大鼠的前列腺湿重、前列腺指数降低,前列腺组织腺上皮及间质面积缩小;并能降低EGF在前列腺组织中的表达,增加TGF-β1的表达。结论:蜣螂乙醇提取物可能通过抑制前列腺组织中睾酮向二氢睾酮转化,调节大鼠雌/雄激素比例平衡,抑制前列腺上皮组织与间质增生,调节EGF与TGF-β1在前列腺组织中的表达,从而抑制前列腺增生。
Objective: To investigate the effect of ethanol extract of dung beetle on rat prostatic hyperplasia and its possible mechanism. Methods: The model of benign prostatic hyperplasia (BPH) was induced by subcutaneous injection of testosterone propionate in ovariectomized male rats. The model rats were divided into three groups: the dung beetle extract, the finasteride solution, the same volume of distilled water, the normal control group and the model Rats were administered with equal volume of distilled water once a day for 4 weeks. After the last administration for 1 h, rats were sacrificed, serum testosterone and estradiol levels and prostate index were measured, and the morphology of prostate was observed. The expression of epidermal growth factor (EGF) and transforming growth factor-β1 (TGF-β1) in rat prostatic tissue was detected by immunohistochemistry. Results: 20g crude drug / kg of dung beetle increased the level of serum testosterone and decreased the level of estradiol in the model rats, and at the same time decreased the wet weight of the prostate and the prostate index and the area of the glandular epithelium and interstitium in the prostate. The expression of EGF in prostate tissue increases the expression of TGF-β1. Conclusion: The ethanol extracts of dung beetle may regulate the ratio of EGF to TGF-β1 in prostate tissue by inhibiting the conversion of testosterone to dihydrotestosterone in prostate tissue, regulating the ratio of estrogen / androgens in rats, inhibiting the proliferation of prostate epithelium and interstitial tissue Expression, thereby inhibiting benign prostatic hyperplasia.
