目的探讨肺癌患者肿瘤组织中层粘连蛋白5(LN5)和表皮生长因子受体(EGFR)表达水平的关系,为EGFR酪氨酸激酶抑制剂(EGFR-TKI)个体化靶向治疗提供依据。方法应用实时定量PCR方法检测了39例肺癌患者肿瘤组织LN5和EGFRmRNA表达水平,并分析二者之间的关系。结果Pearson相关分析提示LN5和EGFR表达水平呈正相关(r=0.541,P<0.0005)。线性回归分析提示EGFR随LN5表达水平的变化而变化,回归方程为:lgEGFR=2.202+0.458lgLN5。结论本研究提示LN5的表达水平和EGFR-TKI的靶分子EGFR的表达水平呈线性相关,LN5的低水平表达可能是肺癌中EGFR-TKI靶向治疗疗效的预测因子之一。本研究为EGFR-TKI的个体化靶向治疗提供了试验依据。
Objective To investigate the relationship between the expression of laminin 5 (LN5) and epidermal growth factor receptor (EGFR) in lung cancer patients and to provide evidence for the targeted therapy of EGFR tyrosine kinase inhibitor (EGFR-TKI). Methods Real-time quantitative PCR was used to detect the expression of LN5 and EGFR mRNA in 39 lung cancer patients and to analyze the relationship between the two. Results Pearson correlation analysis showed that there was a positive correlation between LN5 and EGFR expression (r = 0.541, P <0.0005). Linear regression analysis showed that EGFR changed with the change of LN5 expression, the regression equation was: lgEGFR = 2.202 + 0.458lgLN5. Conclusions This study suggests that the expression level of LN5 is linearly correlated with the expression level of EGFR-TKI EGFR. The low expression of LN5 may be one of the predictors of EGFR-TKI targeted therapy in lung cancer. This study provides the experimental basis for the personalized targeted therapy of EGFR-TKI.
