目的探讨大肠癌中TP的表达与5-Fu化疗敏感性的相关性,评估TP作为5-Fu化疗敏感性预测指标的可行性。方法应用MTT法对体外培养的42例大肠癌进行5-Fu药敏试验,再用免疫组化法对42例大肠癌组织进行TP检测,并将其表达情况与对应的化疗耐药性进行统计学相关分析。结果 TP在大肠癌组织中主要表达于间质巨噬细胞,在癌细胞中的阳性表达率为24.7%,主要表达于癌细胞胞质;TP在癌细胞中的表达与肿瘤组织分级、有无淋巴结转移、Duck’s分期有关(P<0.05),与年龄、性别、组织学类型无关(P>0.05);癌细胞TP阳性表达组与TP阴性表达组对5-Fu敏感率分别为90.0%、9.38%(χ2=9.10,P=0.002),两组差异有统计学意义(P<0.05)。结论检测大肠癌中TP的表达情况对患者选择5-FU化疗有一定的指导意义。
Objective To investigate the relationship between the expression of TP and the sensitivity of 5-Fu chemotherapy in colorectal carcinoma and to evaluate the feasibility of TP as a predictor of 5-Fu chemotherapy sensitivity. Methods 42 cases of colorectal cancer cultured in vitro were subjected to 5-Fu drug sensitivity test by MTT method, and 42 cases of colorectal cancer tissues were examined by immunohistochemical method for the detection of TP, and their expression and corresponding chemoresistance were statistically analyzed Learning correlation analysis. Results TP was mainly expressed in interstitial macrophages in colorectal cancer tissues. The positive expression rate of TP in cancer cells was 24.7%, which was mainly expressed in the cytoplasm of cancer cells. The expression of TP in cancer cells was correlated with tumor grade Lymph node metastasis and Duck’s stage (P <0.05), but not with age, sex and histological type (P> 0.05). The sensitivity of TP-positive cells to TP-positive cells was 90.0%, 9.38 % (χ2 = 9.10, P = 0.002), the difference between the two groups was statistically significant (P <0.05). Conclusion Detecting the expression of TP in colorectal cancer has certain guiding significance for patients choosing 5-FU chemotherapy.
