为证实化疗中血小板因子4(PF4)对正常骨髓细胞的保护作用,用体外培养人骨髓细胞,以MTT法测定细胞的存活率,观察PF4预处理后,骨髓细胞对化疗药物敏感性的变化。经PF4(5μg/ml)预处理48小时后,骨髓细胞对柔红霉素、阿霉素、鬼臼乙叉甙、氨甲喋呤和长春新碱的敏感性降低,药物的半数抑制浓度(IC50)增加(P<0.05),其作用与PF4的浓度及预处理的时间有关。结果提示,PF4能减轻化疗药物对正常人骨髓细胞的细胞毒性,可望成为一种肿瘤化疗时的骨髓保护剂
To confirm the protective effect of platelet factor 4 (PF4) on normal bone marrow cells in chemotherapy, human bone marrow cells were cultured in vitro and the viability of the cells was measured by MTT assay. Changes in susceptibility of bone marrow cells to chemotherapeutic drugs after PF4 pretreatment were observed. After 48 hours of PF4 (5μg/ml) pretreatment, the sensitivity of myeloid cells to daunorubicin, doxorubicin, etoposide, methotrexate and vincristine was reduced, and the IC50 of the drug was increased. (P<0.05), its effect is related to the concentration of PF4 and the time of pretreatment. The results suggest that PF4 can reduce the cytotoxicity of chemotherapeutic drugs on normal human bone marrow cells, and it is expected to be a bone marrow protector for cancer chemotherapy.
