目的 探讨前脑缺血再灌注引起的海马CA_1区神经元死亡是否包含凋亡机制的参与以及缺血预处理(IPC)是否通过影响凋亡抑抑制基因bcl-2的表达起到脑缺血损伤保护作用。方法(1)动物模型建立及分组;选用健康成年雄性Wis-tar大鼠,参照Pulsinelli的四血管阻断法建立前脑缺血模型。动物分为3组;单纯缺血再灌注组(IR组),给子假手术3d后行8min致死性缺血;缺血预处理组(IPC组),先给予3min短暂缺血,3d后再给予8min缺血;对照组,只给予假手术,不给于缺血刺激。(2)结合PI染色的流氏细胞术检测:于缺血再灌注24h。迅速取两背侧新鲜海邓组织,采用流氏细胞仪(FCM)进行海马CA_1区神经元细胞凋亡的检测,低于G_1期DNA含量的细胞为凋亡细胞。(3)免疫组化检测:于缺皿再灌注2h,取脑制备冰冻切片,采用SABC法检测各组海马CA_1区神经元bcl-2及 bcl-xl训蛋白的表达。结果流氏细胞术检测显示,IR组及 IPC组缺血再灌注 24h时海马CA_1区神经元的凋亡百分率分别为13.11±3.00%和2.40±0.98%,与对照组(凋亡百分率为I.34±0.62%)相比,均有显著性差异(P<0.00?
Objective To investigate whether neuronal death in the hippocampal CA1 region caused by forebrain ischemia-reperfusion involves the mechanism of apoptosis and whether ischemic preconditioning (IPC) plays a role in cerebral ischemia injury by affecting the expression of apoptosis-suppressing gene bcl-2 Protective effects. Methods (1) Animal models were established and divided into groups. Healthy adult male Wis-tar rats were used to establish forebrain ischemia model with reference to Pulsinelli’s four-vessel occlusion method. The animals were divided into 3 groups. The ischemia-reperfusion group (IR group) was given 8-month lethal ischemia after sub-sham operation. The ischemic preconditioning group (IPC group) Given 8min ischemia; control group, only given sham surgery, do not give ischemic stimulation. (2) Flow cytometry combined with PI staining: 24h after ischemia-reperfusion. Quickly take fresh dorsal seawater tissue from both sides and detect the apoptosis of hippocampal CA1 neurons by flow cytometry (FCM). The apoptotic cells are lower than the DNA of G_1 phase. (3) Immunohistochemistry: Frozen sections were prepared at 2h after reperfusion, and the expressions of bcl-2 and bcl-xlcr in hippocampal CA1 were detected by SABC method. Results Flow cytometry showed that the apoptotic percentage of hippocampal CA1 neurons was 13.11 ± 3.00% and 2.40 ± 0.98% respectively in IR group and IPC group at 24h, compared with control Group (the percentage of apoptosis was I.34 ± 0.62%), there was a significant difference (P <0.00?
