目的:研究二冬膏对脂多糖(LPS)致急性肺损伤(ALI)模型鼠肿瘤坏死因子-α(TNF-α),白介素-6(IL-6)含量及水通道蛋白5(AQP-5)表达的影响。方法:将72只清洁级SD大鼠随机分为6组:正常组、模型组、二冬膏高、中、低剂量组、地塞米松组,每组12只。除正常组外,其余各组采用一次性尾静脉注射LPS 6 mg·kg-1复制ALI大鼠模型。首次给药在造模后0.5 h,1次/d,连续4 d。二冬膏高、中、低剂量组分别ig二冬膏10,5,2.5 g·kg-1;正常组与模型组ig生理盐水10 m L·kg-1,地塞米松组ip地塞米松0.01 g·kg-1。4 d后处死大鼠,取血清和肺组织检测。采用双抗体夹心-酶联免疫吸附(ABC-ELISA)法检测大鼠血清TNF-α,IL-6的含量,Western bloting测定大鼠肺组织AQP-5蛋白的表达,苏木素和伊红(HE)染色观察肺组织病理形态。结果:与正常组比较,模型组大鼠血清TNF-α,IL-6含量明显升高(P<0.01),肺组织AQP-5蛋白表达水平显著降低(P<0.01)且肺部病理炎症增加。与模型组比较,二冬膏各组均能明显降低TNF-α,IL-6含量(P<0.05,P<0.01),显著升高肺组织AQP-5蛋白的表达(P<0.05,P<0.01)并且减轻肺部病理炎症。结论:二冬膏通过降低ALI模型鼠促炎因子TNF-α,IL-6含量,升高肺组织AQP-5蛋白的表达,改善肺部病理炎症,减轻肺水肿状态,从而有效防治急性肺损伤。
Objective: To investigate the effects of ErDong Ointment on the levels of TNF-α, IL-6 and AQP-5 in acute lung injury induced by lipopolysaccharide (LPS) ) Expression. Methods: Seventy-two clean-grade SD rats were randomly divided into 6 groups: normal group, model group, high-dose, middle-low dose group and dexamethasone group, 12 rats in each group. Except for the normal group, the rats in each group were injected with LPS 6 mg · kg-1 once a single intravenous injection into the ALI rat model. The first administration in 0.5 h after modeling, 1 time / d, for 4 days. Two winter ointment high, medium and low dose groups were ig two winter paste ointment 10,5,2.5 g · kg-1; normal group and model group ig saline 10 m L · kg-1, dexamethasone group ip dexamethasone 0.01 g · kg-1.4 d after the rats were sacrificed, serum and lung tissue were taken for detection. Serum levels of TNF-α and IL-6 were detected by double antibody sandwich enzyme-linked immunosorbent assay (ABC-ELISA). The expression of AQP-5 protein in lung tissue was detected by Western bloting. The expression of hematoxylin and eosin Staining to observe the histopathology of lung tissue. Results: Compared with normal group, the levels of serum TNF-α and IL-6 in model group were significantly increased (P <0.01), the expression of AQP-5 protein in lung tissue was significantly decreased (P <0.01) . Compared with model group, Shuangdong cream could significantly decrease the contents of TNF-α and IL-6 (P <0.05, P <0.01), and significantly increase the expression of AQP-5 protein in lung tissue 0.01) and reduce lung pathological inflammation. CONCLUSION: Shuangdong ointment can effectively prevent and treat acute lung injury by decreasing the levels of TNF-α and IL-6 and increasing the expression of AQP-5 protein in lung tissue of ALI model rats, improving the pathological inflammation of the lungs and relieving the pulmonary edema state .
