目的 测定神经断端套接后 4周内 ,再生室收集液 (regenerationchamber,RCF)中神经生长因子 (nervegrowthfactor,NGF)的含量变化。方法 SD大鼠 3 2只 ,双侧坐骨神经切断后 ,用硅胶再生室套接 ,分别于术后 2 4h、1周、2周、4周各时间点取材。用标准双抗体夹心酶联免疫方法测定收集到的RCF中NGF的含量。结果 四个时间点收集的RCF中NGF的含量分别为 5 5 0pg ml、890pg ml、670pg ml及 40 0pg ml。结论 NGF可能是RCF中发挥神经营养和趋化作用的重要成分之一。
Objective To determine the changes of nerve growth factor (NGF) in regenerative chamber (RCF) within 4 weeks after nerve end-to-end sheathing. Methods Thirty two SD rats were sciatic nerves cut off and sutured with silica gel regeneration chamber. The rats were drawn at 24 hours, 1 week, 2 weeks and 4 weeks after operation respectively. The level of NGF in the collected RCFs was determined by standard double antibody sandwich enzyme-linked immunosorbent assay. Results The levels of NGF in RCF collected at four time points were respectively 550 μg ml, 890 pg ml, 670 pg ml and 40 0 pg ml. Conclusion NGF may be one of the important components of RCF that plays a neurotrophic and chemotactic role.
