目的:探讨内皮抑素质粒联合顺铂对上皮性卵巢癌细胞株A2780增殖和凋亡的影响。方法:通过瞬时转染内皮抑素质粒到卵巢癌细胞A2780后,采用MTT法和流式细胞术检测内皮抑素以及内皮抑素和顺铂联合应用对细胞增殖和凋亡的影响。结果:MTT法检测发现转染内皮抑素质粒后,A2780细胞的增殖明显减慢,顺铂对其生长抑制作用明显增强,差异有统计学意义(P<0.05)。流式细胞仪检测发现与空白组相比,转染内皮抑素质粒后A2780细胞的G0/G1期细胞明显增加,S期细胞明显减少,细胞的凋亡率由(1.59±0.86)%增加到(8.66±1.27)%;内皮抑素质粒联合顺铂治疗后,上述差异更加显著(P<0.01)。结论:内皮抑素联合顺铂可通过抑制A2780细胞的生长和增加其凋亡而产生协同抗瘤作用。
Objective: To investigate the effect of endostatin plasmid combined with cisplatin on the proliferation and apoptosis of epithelial ovarian cancer cell line A2780. Methods: After transient transfection of endostatin plasmid into ovarian cancer cell line A2780, MTT assay and flow cytometry were used to detect the effects of endostatin and endostatin combined with cisplatin on cell proliferation and apoptosis. Results: MTT assay showed that the proliferation of A2780 cells was obviously slowed down after transfected with endostatin plasmids, and the inhibitory effect of cisplatin on its growth was significantly enhanced (P <0.05). Flow cytometry showed that compared with the blank group, A2780 cells transfected with endostatin plasmid G0 / G1 phase cells increased significantly, S phase cells decreased significantly, the cell apoptosis rate increased from (1.59 ± 0.86)% to (8.66 ± 1.27)% respectively. The difference between the two groups was more significant (P <0.01) after treatment with endostatin plasmid and cisplatin. Conclusion: Endostatin plus cisplatin can produce synergistic antitumor effect by inhibiting the growth of A2780 cells and increasing their apoptosis.
