目的优选PCO大鼠的造模方法。方法分别采用来曲唑、硫酸普拉睾酮钠、硫酸普拉睾酮钠联合HCG诱导PCO大鼠模型。应用免疫组化法检测MMP-9、TIMP-1在大鼠卵巢组织中的表达,HE染色后光镜下观察卵巢局部形态。结果 MMP-9在模型组大鼠卵巢组织中的表达增高,差异有显著统计学意义(P<0.01);TIMP-1表达在模型组大鼠卵巢组织中均降低,差异有统计学意义(P<0.01,P<0.05)。3种造模方法均能诱导PCO动物模型,其中来曲唑造模法最优,符合PCOS动物造模要求。结论采用来曲唑造模法诱导PCO大鼠动物模型效果最佳。
Objective To optimize the modeling method of PCO rats. Methods Letrozole, sodium pramipexole sulfate, sodium pramipetimate and HCG were used to induce PCO rat model. Immunohistochemistry was used to detect the expression of MMP-9 and TIMP-1 in rat ovarian tissue. The morphology of ovary was observed under light microscope after HE staining. Results The expression of MMP-9 in the model group was significantly higher than that in the model group (P <0.01). The expression of TIMP-1 in the model group was significantly lower than that in the model group (P <0.01, P <0.05). Three kinds of modeling methods can induce PCO animal models, of which Letrozole modeling method is the best, in line with PCOS animal modeling requirements. Conclusion Letrozole modeling induced the best animal model in PCO rats.
