目的:探讨饲料锌对腹腔巨噬细胞(Peritonealmacrophage,PMΦ)吞噬功能的影响及其机理。方法:建立大鼠缺锌、高锌模型,测定大鼠PMΦ吞噬率、吞噬指数及cAMP、cGMP。结果:①缺锌(ZD)组PMΦ吞噬率、吞噬指数均非常显著地低于配对(PF)组及自由喂养(AL)组,经补锌后得以恢复;高锌(ZE)组明显低于AL组。②PMΦ经体外脂多糖(LPS)作用后胞内cAMP含量、cAMP/cGMP水平ZD组明显低于PF、AL及缺锌补锌(ZD+AL)组;ZE组非常显著地高于AL组。结论:不适量锌可能通过PMΦ胞内cAMP/cGMP变化而影响PMΦ活化,进而抑制其吞噬杀菌功能。
Objective: To investigate the effects of dietary zinc on the phagocytic function of peritoneal macrophage (PMΦ) and its mechanism. Methods: Zinc-deficient and high-zinc model rats were established. The phagocytic rate, phagocytic index and cAMP, cGMP of rats were measured. RESULTS: ① The phagocytic rate and phagocytic index of PMΦ in ZD group were significantly lower than those in PF group and AL group, and were restored after Zinc supplementation. Zinc group was significantly lower than that in ZE group AL group. (2) The intracellular cAMP level and cAMP / cGMP level in PMΦ treated by lipopolysaccharide (LPS) in vitro were significantly lower than that in PF, AL and ZD + AL groups; ZE group was significantly higher than that in AL group. Conclusion: Zinc deficiency may affect the activation of PMΦ by PMΦ intracellular cAMP / cGMP changes, thereby inhibiting its phagocytosis.
