目的探讨脂肪源性干细胞(ADSCs)对大鼠阴道黏膜下生物补片生物相容性的影响。方法 2010年4月至2011年2月在广州医学院第一附属医院,获取大鼠腹股沟处脂肪组织分离培养ADSCs,选择第3代细胞与脱细胞猪心包膜(APP)、脱细胞牛心包膜(ABP)两种生物补片构建新型组织工程补片,选取体外培养1、7d的新型组织工程补片进行扫描电镜。根据大鼠阴道黏膜下植入生物补片的不同类型,将20只去势SD雌性大鼠随机分为5组:APP组,ABP组,APP+ADSCs组,ABP+ADSCs组,对照组。术后30d植入部位取材,HE染色观察组织的炎症反应程度,巨噬细胞及异物巨细胞计数。结果 ADSCs可以在两种生物补片上黏附及生长。与ABP组比较,APP组局部的炎症反应及异物反应较重,补片结构破坏明显。加载ADSCs的两种生物补片植入局部组织的炎症反应及异物反应明显低于未加载ADSCs的生物补片。结论 ABP较APP具有更好的生物相容性。ADSCs可以调节生物补片植入部位的炎症反应及异物反应,可能对生物补片的降解有缓解作用。
Objective To investigate the effect of adipose derived stem cells (ADSCs) on biocompatibility of vaginal submucosal bioprostheses in rats. Methods From April 2010 to February 2011, ADSCs were isolated and cultured in the inguinal fat tissue of rats in the First Affiliated Hospital of Guangzhou Medical College from April 2010 to February 2011, and the third generation cells were selected for combination with acellular porcine apocrine membrane (APP) (ABP) two kinds of bio-patch to build a new tissue engineering patch, choose a new tissue engineering patch 1,7d in vitro scanning electron microscopy. Twenty female SD rats were randomly divided into 5 groups: APP group, ABP group, APP + ADSCs group, ABP + ADSCs group and control group according to the different types of vaginal submucosal implanted biological patch. The site of implantation was taken 30 days after operation. The degree of inflammatory reaction, the number of macrophages and foreign body giant cells were observed by HE staining. Results ADSCs can adhere and grow on two bio-patches. Compared with the ABP group, APP group local inflammatory reaction and foreign body reaction heavier, obvious patch structure destruction. Local biofilms implanted with ADSCs implanted in the local tissue showed significantly lower inflammatory and foreign body reaction than those without ADSCs. Conclusion ABP has better biocompatibility than APP. ADSCs can modulate the inflammatory response and foreign body reaction at the implant site of biological patch, which may alleviate the degradation of biological patch.
