目的了解2007-2008年间吉林省H3N2亚型流感病毒HA1基因的演变特征。方法采用MD-CK细胞分离培养流感病毒,提取病毒RNA,经逆转录和聚合酶链式反应扩增后测序。结果①2007-2008年吉林省流行的H3N2亚型流感毒株核苷酸同源性为98.9%~100%,与同期WHO推荐的疫苗株A/Wisconsin/67/05相比有10~14个氨基酸位点发生变化,并且在122位增加了一个糖基化位点;②长春市11月与12月分离的病毒株在进化树上处于不同的两个侧枝;③在同一流行高峰吉林省8个市州分离的流感病毒株在进化树上形成不同的两簇。结论2007-2008年吉林省流行的H3N2亚型流感毒株与WHO推荐的疫苗株A/Wisconsin/67/05相比已经发生一定变化;不同时间及市州流行的病毒株也有差异。
Objective To understand the evolution of HA1 gene of H3N2 influenza virus in Jilin province from 2007 to 2008. Methods The influenza virus was isolated and cultured by MD-CK cells, and the viral RNA was extracted and amplified by reverse transcription and polymerase chain reaction (PCR). Results ① The homology of H3N2 influenza virus strains in Jilin province between 2007 and 2008 was 98.9% ~ 100%, which was 10 ~ 14 amino acids compared with WHO recommended vaccine strain A / Wisconsin / 67/05 The site changed, and a glycosylation site was added at 122. ② The strains isolated in November and December in Changchun were in two different branches in the phylogenetic tree; ③ In the same epidemic peak, 8 in Jilin Province The influenza strains isolated in the city-states form two different clusters in the evolutionary tree. Conclusion The prevalence of H3N2 subtype influenza viruses in Jilin Province during 2007-2008 has changed a little compared with the WHO recommended vaccine strain A / Wisconsin / 67/05. There are also differences in the virus strains prevailing at different times and in cities and prefectures.
