目的探讨人肾癌细胞核因子-κB p65(NF-κB p65)表达与微血管生成的关系。方法免疫组化染色法检测40例肾癌组手术切除的肾癌细胞NF-κB p65表达,通过标记CD34阳性表达检测微血管密度(MVD)。对不同分期肾癌细胞内NF-κB p65阳性表达率及MVD进行比较并分析其与患者生存期关系。对照组10例肾组织取自远离肾癌的正常肾组织。结果 40例肾癌组织中,26例肾癌细胞内NF-κB p65阳性表达,对照组肾组织中未见阳性表达(P<0.01)。其中,晚期肾癌细胞内NF-κB p65阳性表达率明显高于肾癌早期(P<0.05)。与肾癌细胞内NF-κB p65阴性表达者比较,肾癌细胞NF-κB p65阳性表达者MVD数明显升高(P<0.01),而患者5年生存率明显降低(P<0.05)。结论肾癌细胞内NF-κB p65阳性表达与微血管生成密切相关,影响患者病情发展及预后。
Objective To investigate the relationship between the expression of nuclear factor-κB p65 (NF-κB p65) and micro-angiogenesis in human renal cell carcinoma. Methods Immunohistochemical staining was used to detect the expression of NF-κB p65 in 40 cases of renal cell carcinoma. The expression of CD34 was detected by microvessel density (MVD). The positive expression rate of NF-κB p65 and the MVD in different stages of renal cell carcinoma were compared and their relationship with the survival time was analyzed. Ten patients in the control group were taken from normal kidney tissue far from renal cell carcinoma. Results In 40 cases of renal cell carcinoma, 26 cases of renal cell carcinoma showed positive expression of NF-κB p65, but no positive expression in the control group (P <0.01). Among them, the positive rate of NF-κB p65 in advanced renal cell carcinoma was significantly higher than that in early stage of renal cell carcinoma (P <0.05). Compared with the negative expression of NF-κB p65 in renal cell carcinoma, the MVD of NF-κB p65 in renal cell carcinoma was significantly increased (P <0.01), while the 5-year survival rate of renal cell carcinoma was significantly decreased (P <0.05). Conclusion The positive expression of NF-κB p65 in renal cell carcinoma is closely related to angiogenesis, which affects the progression of the disease and prognosis.
