目的研究HOXD10基因在人脑胶质瘤组织中的表达及其意义。方法收集具有明确病理分级的48例脑胶质瘤和8例正常脑组织标本,应用RT-PCR和Western blot方法检测HOXD10在不同级别人脑胶质瘤和正常脑组织中HOXD10基因mRNA和蛋白水平的表达,并进一步研究了HOXD10与胶质瘤病理分级的相关性。结果 HOXD10在正常脑组织中呈高表达。Ⅲ~Ⅳ级胶质瘤组织中的HOXD10基因mRNA和蛋白表达明显低于Ⅰ~Ⅱ级。结论 HOXD10在正常脑组织中呈高表达,并与脑胶质瘤恶性程度呈负相关。
Objective To study the expression of HOXD10 gene in human glioma and its significance. Methods 48 cases of glioma and 8 cases of normal brain tissue with definite histological grade were collected. HOXD10 mRNA and protein levels were detected by RT-PCR and Western blot in different grades of human glioma and normal brain tissue And further study the correlation between HOXD10 and pathological grade of glioma. Results HOXD10 was highly expressed in normal brain tissue. The mRNA and protein expressions of HOXD10 in grade Ⅲ ~ Ⅳ glioma tissues were significantly lower than those in grade Ⅰ ~ Ⅱ. Conclusion HOXD10 is highly expressed in normal brain tissue and negatively correlated with the malignant degree of glioma.
