应用细胞培养和分子截留技术,通过体内和体外实验,观察了不同浓度葡萄糖对大鼠腹膜间皮细胞和腹腔巨噬细胞产生白细胞介素-6和巨噬细胞吞噬功能的影响。结果表明,应用1.5%和4.25%浓度的葡萄糖腹膜透析液给大鼠腹腔灌注3周后,腹膜间皮细胞和腹腔巨噬细胞产生白细胞介素-6的能力明显下降,体外实验也证实了这一点,同时发现,1.5%和4.25%浓度的葡萄糖亦明显抑制了巨噬细胞的吞噬功能,尤其以4.25%浓度的葡萄糖更为明显。由此可见,腹透液葡萄糖浓度越高,影响腹膜间皮细胞和巨噬细胞的免疫和防御功能越明显,从而更易于使腹膜透析患者并发腹膜感染。
The effects of different concentrations of glucose on phagocytosis of interleukin-6 and macrophage in peritoneal mesothelial cells and peritoneal macrophages were observed by in vitro and in vivo experiments with cell culture and molecular entrapment techniques. The results showed that the ability of peritoneal mesothelial cells and peritoneal macrophages to produce interleukin-6 significantly decreased after peritoneal perfusion with 1.5% and 4.25% glucose peritoneal dialysate for 3 weeks. In vitro experiments Also confirmed this point, also found that 1.5% and 4.25% of the concentration of glucose also significantly inhibited the phagocytosis of macrophages, especially with 4.25% glucose concentration is more obvious. Thus, the higher density of peritoneal dialysate glucose, the more obvious the immune and defense functions affecting peritoneal mesothelial cells and macrophages, making it easier for peritoneal dialysis patients complicated with peritoneal infection.
