目的 建立和优化蛋白质分子差异展示的双向聚丙烯酰胺凝胶电泳技术 ,初步观察番泻叶提取物诱导小鼠结肠组织中蛋白质的差异表达 ,为进一步筛选其中介导番泻叶生物作用的蛋白质分子奠定基础 .方法 应用番泻叶提取物灌胃制备小鼠慢性胃肠运动增强模型 ,提取其结肠组织中总蛋白质 ,优化蛋白质分子差异展示的双向聚丙烯酰胺凝胶电泳技术 ,并用其分析模型动物结肠组织中蛋白质 ,双向电泳凝胶银染法显示其蛋白质的差异表达 .结果 建立了小鼠慢性胃肠运动增强模型 ,优化了双向聚丙烯酰胺凝胶电技术并用其展示出模型动物结肠组织中有差异蛋白质的表达 .结论 番泻叶提取物可以诱导小鼠结肠组织中蛋白质差异表达 ,这些差异蛋白质可能介导了番泻叶的生物作用
Objective To establish and optimize two-dimensional polyacrylamide gel electrophoresis for protein differential display, and to observe the differential expression of proteins in mice’s colon tissue induced by senna extract to further screen protein molecules that mediate the biological effects of senna. To establish the basis. Methods To apply senna extract to gavage to establish a mouse model of chronic gastrointestinal motility enhancement, extract the total protein in the colon tissue, optimize the two-dimensional polyacrylamide gel electrophoresis technique for protein differential display, and use it to analyze model animals. Proteins in colon tissues were differentially expressed by two-dimensional electrophoresis gel silver staining. Results The mouse chronic gastrointestinal motility enhancement model was established. The bidirectional polyacrylamide gel electroporation technique was optimized and used to display the colon tissue of the model animals. There are differential protein expressions. Conclusion The senna extract can induce differential expression of proteins in the mouse colon. These differential proteins may mediate the biological effects of Senna.
