Observing the chromosome of Drosophila salivary gland cells is a major experiment in high school biology. Through experiments, students have a perceptual understanding of chromosomes, the carrier of genetic material. Although in many publications, the production method of the fruit fly flesh gland salivary glands has been described in detail, many middle school teachers still find it difficult. The main problem is that the chromosomes are clustered and the arms of the chromosomes are not stretched, affecting the experimental results. To this end, we conducted a variety of experimental explorations to better effect the cryogenic freezing method of third-instar larvae.
