利福布丁(rifabutin)是利福平S的半合成衍生物,具有广谱抗菌活性,其主要代谢产物为25-去乙酰基衍生物LM-565。本文报道用HPLC-UV法测定血浆及尿液中利福布丁及LM-565。色谱条件:Ultrasphere ODS反相柱(25cm×4.6mm,5μm),柱温40℃,流动相为乙腈-0.05 mol/L磷酸钾(pH 4.2)-三乙胺(38:61.5:0.5,v/v),流速为1.0ml/min,吸收波长为275 nm,内标为美达西泮(5ng/ml)甲醇溶液。标准品及对照品制备:在棕色容量瓶中配制利福布丁和LM-565甲醇储备液,于4℃
Rifabutin is a semisynthetic derivative of rifampicin S with broad spectrum antibacterial activity. Its main metabolite is 25-deacetyl derivative LM-565. This paper reports the determination of rifabutin and LM-565 in plasma and urine by HPLC-UV. Chromatographic conditions: Ultrasphere ODS reverse phase column (25cm × 4.6mm, 5μm) with a column temperature of 40 ℃ and mobile phase of acetonitrile-0.05mol / L potassium phosphate at pH 4.2-triethylamine (38: 61.5: v) at a flow rate of 1.0 ml / min and an absorption wavelength of 275 nm with an internal standard of 5 mg / ml in methanol.
