目的通过建立动物的近视眼模型,观察近视眼发展期和恢复期屈光状态的改变以及基质金属蛋白酶(MMP-2)的含量变化,探讨小鸡近视眼模型巩膜重塑过程中的生物化学变化机制。设计随机对照实验性研究。研究对象32只2天龄SPF美国进口白种莱航鸡。方法将32只鸡随机分为A、B、C、D四组,每组8只,A、B组为实验组,用透明眼罩遮盖小鸡的右眼进行单眼形觉视觉剥夺,左眼为对照;C、D组为正常对照。21天后去遮盖,检测双眼屈光状态和眼轴,处死A组和C组并分离7mm直径的后巩膜组织;继续饲养B组和D组至去遮盖后21天,检测双眼屈光状态和眼轴后处死并分离7mm直径的后巩膜组织。用酶活性分析法测定所有动物巩膜中MMP-2的含量,并将实验组和对照组的数据进行统计学分析。主要指标遮盖后及去遮盖后小鸡的屈光度、眼轴,巩膜中MMP-2的含量。结果遮盖21天A、B组左右眼屈光度比较,差异有统计学意义(P= 0.000),平均相差(-28.38±6.72)D;A组左右眼眼轴相比差异有统计学意义(P=0.000),平均相差(1.47±0.37)mm;遮盖眼后极部巩膜中的MMP-2含量显著性升高,A组遮盖眼与对侧无遮盖眼间比较差异有统计学意义,平均增加(76.00±15.20)%。去遮盖21天B组左右眼屈光度比较差异无统计学意义,平均差值(0.19±0.92)D;B组左右眼眼轴比较差异无统计学意义,平均差值(0.15±0.50)mm;去遮盖眼后极部巩膜中的MMP-2含量明显下降,与对照组之间差异无统计学意义(P=0.47)。结论形觉剥夺性近视眼的小鸡模型中,近视眼的形成与后巩膜中的MMP-2水平有关,而且MMP-2的含量在近视眼发展期和恢复期存在一定的变化规律。(眼科,2006,15:334-337)
OBJECTIVE: To establish a model of myopic eye in rats and observe the changes of refractive status and the content of matrix metalloproteinase (MMP-2) during the development and recovery of myopia, and to explore the biochemical changes in scleral remodeling of myopic chick model mechanism. Design Randomized controlled experimental study. Study objects 32 2-day-old SPF United States imported white Lai Lai chicken. Methods Thirty-two chickens were randomly divided into four groups (A, B, C and D), with 8 rats in each group. Groups A and B were experimental groups. The eyes of the chickens were covered with a transparent eye mask and visual deprived by one eye. Control; C, D group as a normal control. After 21 days, the eyes were examined for refractive status and axial length of the eyes. A and C groups were sacrificed and 7 mm diameter posterior scleral tissues were removed.
