目的:研究印记基因Neuronatin(NNAT)在人体常见肿瘤中的表达情况及其两种编码产物NNATα和NNATβ对肿瘤细胞增殖的影响。方法:运用组织芯片免疫组化技术对一些人体常见肿瘤及其对应正常组织中NNAT的表达进行系统检测;同时利用腺病毒载体技术,将NNATα和NNATβ分别导入人结肠癌细胞系SW620,并用实时细胞分析仪和平板克隆实验分析细胞增殖能力的变化。结果:①免疫组化结果显示:NNAT在食道鳞癌,结肠腺癌,直肠腺癌,胰腺腺癌,乳腺非特殊性浸润性导管癌,宫颈鳞癌,子宫内膜癌,膀胱移行上皮癌,肺鳞癌,皮肤鳞癌以及前列腺腺癌11种肿瘤组织,肾上腺,皮肤,睾丸,脑,骨骼肌,胰腺6种正常人体组织,以及慢性结肠黏膜炎与慢性肝炎2种炎性组织中呈阳性。②体外细胞增殖实验结果显示,NNATα对人结肠癌肿瘤细胞增殖有一定的抑制作用。结论:NNAT基因在多种人体肿瘤组织中表达,其α片段的表达对于结肠癌细胞系SW620增殖具有轻微抑制作用。
Objective: To study the expression of Neuronatin (NNAT) in human common tumors and the effects of two coding products, NNATα and NNATβ, on the proliferation of tumor cells. Methods: The expression of NNAT in some common tumors and corresponding normal tissues was systematically detected by tissue microarray immunohistochemistry. At the same time, NNATα and NNATβ were respectively introduced into human colon cancer cell line SW620 by using adenovirus vector technique. Analyzer and plate cloning experiments to analyze changes in cell proliferation. Results ① Immunohistochemical results showed that: NNAT in esophageal squamous cell carcinoma, colon adenocarcinoma, rectal adenocarcinoma, pancreatic adenocarcinoma, breast non-specific invasive ductal carcinoma, cervical squamous cell carcinoma, endometrial carcinoma, bladder transitional cell carcinoma, Lung squamous cell carcinoma, squamous cell carcinoma of the lung, and prostate adenocarcinoma of 11 kinds of tumor tissue, adrenal gland, skin, testis, brain, skeletal muscle and pancreas, and 6 kinds of normal human tissues of the pancreas, as well as two inflammatory tissues of chronic colonic mucositis and chronic hepatitis . ② In vitro cell proliferation experiments showed that NNATα inhibited the proliferation of human colon cancer cells to a certain extent. Conclusion: The NNAT gene is expressed in a variety of human tumor tissues. The expression of α fragment has a slight inhibitory effect on the proliferation of colon cancer cell line SW620.
