目的研究渗透促进剂对蛇床子素的体外经皮渗透的影响。方法将蛇床子有效部位制成饱和生理盐水溶液,采用Valia-Chien扩散池考察了氮酮、薄荷醇及土荆芥油3种经皮吸收促进剂对蛇床子素经离体鼠皮透过性的影响。结果应用土荆芥油、薄荷醇和氮酮为经皮吸收促进剂,药物的稳态流量与对照组比较均有提高,透过倍数分别为3.09、3.41及4.38;药物的扩散系数分别提高1.10、2.18及6.30倍,但分配系数却分别降低1/3、1/3及2/5。结论 3种渗透促进剂的作用机理主要为改变角质层的通透性,减低了药物经皮肤透过的阻力,提高了药物在皮肤角质层的扩散系数。
Objective To study the effect of penetration enhancers on percutaneous osmosis of osthole. Methods The effective part of Cnidium monnieri was made into saturated saline solution. The effects of Azone, menthol and cathartane oil on transdermal percutaneous permeation of osthole were investigated using Valia-Chien diffusion cell. Impact. Results The soil catarrhal oil, menthol and azone were used as percutaneous absorption enhancer. The steady-state flow rate of the drug increased compared with the control group, the permeation times were 3.09, 3.41 and 4.38, respectively. The diffusion coefficient of drug increased by 1.10, 2.18 and 6.30 times respectively, but the distribution coefficients were reduced by 1/3, 1/3 and 2/5 respectively. Conclusion The mechanism of the three penetration enhancers is mainly to change the permeability of the stratum corneum, reduce the resistance of the drug through the skin, and improve the diffusion coefficient of the drug in the stratum corneum of the skin.
