[目的]研究化学转化过程中热激后冰浴时间和复苏时间对转化效率的影响,确定简便快速的转化方法。[方法]采用2种缓冲溶液制备感受态,以热激后冰浴时间或复苏时间为变量,研究这2个因素与转化效率的关系。[结果]冰浴2min,复苏30或40min达到最佳转化效率;冰浴和复苏时间为0时仍能获得相当数量的转化子。[结论]热激后冰浴和复苏时间对转化效率有一定影响,但不是决定性因素。因此,在一般性转化试验中可以省略这2个步骤以节省时间。
[Objective] The research aimed to study the influence of ice bath time and recovery time on conversion efficiency after heat shock in chemical conversion process, and to determine a simple and rapid conversion method. [Method] Two kinds of buffer solutions were used to prepare competent cells. The time of ice bath or recovery time was taken as the variables to study the relationship between these two factors and transformation efficiency. [Result] The optimal conversion efficiency was achieved after ice bath for 2 min, recovery for 30 or 40 min, and a considerable number of transformants were obtained when the ice bath and recovery time were zero. [Conclusion] After heat shock, the ice bath and the recovery time have a certain impact on the conversion efficiency, but not the decisive factor. Therefore, these two steps can be omitted in the general transformation experiment to save time.
