目的分析卵巢上皮癌细胞生物学特性受TIZ基因过表达的影响。方法利用脂质体砖染法测定TIZ mRNA表达,采用集落形成实验、细胞体外侵袭重建基膜实验、Transwell小室实验、体外黏附实验检测细胞的各种能力。结果 pc DNA3.1/TIZ-HO9801细胞集落形成率低于阴性对照组及空白对照组,差异有统计学意义(P<0.05)。3组细胞体外迁移、侵袭和黏附能力比较,差异无统计学意义(P>0.05)。结论 TIZ基因过表达会抑制卵巢上皮癌细胞的增殖能力。
Objective To analyze the biological characteristics of ovarian epithelial carcinoma cells under the influence of TIZ gene overexpression. Methods TIZ mRNA expression was determined by liposome brick staining. The colony formation assay, in vitro invasion and rebuilding basement membrane assay, Transwell chamber assay and in vitro adhesion assay were used to detect the cell viability. Results The rate of pcDNA3.1 / TIZ-HO9801 cell colony formation was lower than that of negative control group and blank control group (P <0.05). There was no significant difference in migration, invasion and adhesion among 3 groups of cells (P> 0.05). Conclusion TIZ gene overexpression can inhibit the proliferation of ovarian epithelial cancer cells.
