背景胸痹通胶囊可增加冠心病患者心肌缺血区的血液灌流,减小心肌梗死面积,改善心电图缺血性S-T改变。目的观察胸痹通胶囊对家兔动脉壁一氧化氮代谢的影响,探讨该药改善心肌血液灌流、发挥心肌保护作用。设计随机分组设计,对照实验。单位山东省潍坊市中医院内一科材料实验于2003-04/2004-02在潍坊医学院生理教研室完成。选用健康成年家兔30只,雌雄各半。随机将动物分为3组正常对照组,模型组,治疗组,每组3只。方法①高脂血症动物模型制备正常对照组喂饲基础颗粒饲料,模型组喂饲造模饲料(基础颗粒饲料+3%猪油+1%胆固醇粉),治疗组除喂造模饲料外,第6周末开始每天用胸痹通胶囊(组方太子参、云苓、瓜蒌、清夏、橘红、石菖蒲、郁金、炙复花、降香、当归、麦冬、五味子)按1粒/kg的剂量加生理盐水10m L灌胃3周。共干预8周。②一氧化氮合酶和一氧化氮代谢产物试剂盒由军事医学科学院放射医学研究所提供,按说明书操作。计算一氧化氮合酶活力,以每分钟每克组织产生的一氧化氮量(nm ol)表示活力;用分光光度计在545nm处测样品光密度,亚硝酸钠溶液制作标准曲线,以μmol/L表示血清一氧化氮代谢产物NO2-,NO3-的浓度。③计量资料组间差异比较采用t检验。主要观察指标各组家兔血管壁组织一氧化氮合酶活力和血清一氧化氮代谢产物浓度比较。结果家兔30只均进入结果分析。家兔动脉壁组织一氧化氮合酶活力及血清一氧化氮代谢产物浓度模型组和治疗组均明显低于正常对照组(P<0.05~0.01),治疗组明显高于模型组(P<0.05)。结论①高脂血症家兔动脉壁一氧化氮系统受损害,血清一氧化氮代谢产物水平降低。②胸痹通胶囊治疗可改善上述状况,起到抗心肌缺血、发挥心肌保护的作用。
BACKGROUND: Xiongbitong Capsule can increase blood perfusion in myocardial ischemic region in patients with coronary heart disease, reduce myocardial infarct size, and improve electrocardiographic ischemic S-T changes. Objective To observe the effect of Xiongbitong Capsule on the metabolism of nitric oxide in the arterial wall of rabbits, and to explore the effect of this drug on myocardial hemoperfusion and myocardial protection. Design random grouping design, control experiment. The material experiment of the first department of the Chinese Academy of Internal Medicine, Weifang City, Shandong Province, was completed at the Department of Physiology, Weifang Medical College from April 2003 to February 2004. Thirty healthy adult rabbits were selected, male and female. Animals were randomly divided into 3 groups: normal control group, model group, and treatment group, 3 in each group. Method 1 Preparation of animal model of hyperlipidemia The normal control group was fed with basic pellet feed, the model group was fed with model feed (basal pellet feed + 3% lard + 1 % cholesterol powder), and the treatment group was fed with feed, in addition to feed. On the 6th weekend, start daily with Xiongqitong capsule (group of Pseudostellaria, Yunxiao, Gualou, Qingxia, Tangerine, Shichangpu, Yujin, Zhifuhua, Dangxiang, Angelica, Ophiopogon japonicus, Schisandra) by 1 capsule. The dose of /kg plus normal saline 10m L was given for 3 weeks. A total of 8 weeks intervention. 2 Nitric oxide synthase and nitric oxide metabolite kits were provided by the Institute of Radiation Medicine, Academy of Military Medical Sciences, and were operated according to the instructions. The activity of nitric oxide synthase was calculated. The activity was expressed as the amount of nitric oxide (nm ol) produced per minute per gram of tissue. The optical density of the sample was measured with a spectrophotometer at 545 nm. A standard curve was prepared with a sodium nitrite solution and expressed as μmol/mL. L represents the concentration of nitric oxide metabolites NO2-, NO3-. 3 Comparison of differences in measurement data groups using t test. Nitric oxide synthase activity and serum nitric oxide metabolite concentrations in rabbits’ vascular wall tissue were compared in each group. Results Thirty rabbits were involved in the result analysis. Nitric oxide synthase activity and serum nitric oxide metabolite concentration in the rabbit model group and treatment group were significantly lower than those in the normal control group (P<0.05-0.01), and the treatment group was significantly higher than the model group (P<0.05). ). Conclusion 1 The nitric oxide system in arterial wall of hyperlipidemic rabbits is damaged, and the level of serum nitric oxide metabolites is decreased. 2 Xiongbitong capsule treatment can improve the above conditions, play an anti-myocardial ischemia, play a role in myocardial protection.
