文献中常有介绍,用标记的胸苷(TdR)或尿苷(UR)参入试验判断某种药物对DNA或RNA合成的影响。当TdR或UR参入被抑制时,一般认为DNA或RNA合成受抑制。我们在研究血卟啉衍生物(HpD)对癌细胞的光动力作用时,曾观察到它对~3H-TdR参入有很强的抑制作用,但主要是阻止~3H-TdR在细胞膜的转运,并不反映对DNA合成的抑制。本文应用两种类型抗癌药物(5-FU和氮芥)与HpD对比,观察其对~3H-TdR参入过程的影响,并对HpD的光动力效应作进一步分析。
Commonly described in the literature, the use of labeled thymidine (TdR) or uridine (UR) into the test to determine the effect of a drug on DNA or RNA synthesis. We studied the hematoporphyrin derivative (HpD) on the photodynamic effect of cancer cells, it has been observed that ~ 3H-TdR incorporation has a strong inhibitory effect, but mainly to prevent ~ 3H-TdR in the cell membrane transport, Does not reflect the inhibition of DNA synthesis. In this paper, we compared the effect of 3H-TdR incorporation with two types of anti-cancer drugs (5-FU and nitrogen mustard) and HpD, and further analyzed the photodynamic effect of HpD.
