目的:观察水芹提取物对应激致小鼠慢性疲劳综合征(CFS)的对抗作用及其相关机制。方法:采用强制冷水游泳应激法制备CFS模型,从应激角度,观察应激导致动物疲劳时,其大体行为学、下丘脑—垂体—肾上腺轴(HPA轴)功能变化、中枢神经递质的变化以及骨骼肌能量代谢指标,并进一步探讨水芹提取物治疗慢性疲劳的机制。结果:冷水应激使小鼠自主活动明显下降;HPA轴功能指标:血清促肾上腺皮质激素(ACTH)、皮质醇(CORT)明显升高,性激素睾酮(T)水平下降;中枢递质指标:5-羟色胺(5-HT)、多巴胺(DA)明显上升;外周疲劳骨骼肌能量代谢指标:超微量Ca2+-Mg2+-ATP酶活力降低、磷酸(Pi)含量明显升高,体内自由基水平丙二醛(MDA)明显升高、超氧化物歧化酶活力(SOD)明显下降。药物治疗后,小鼠自主活动能力明显增加、HPA轴与性腺轴功能恢复正常,中枢神经类递质降低,外周疲劳情况好转,体内自由基消除到正常水平。结论:水芹提取物对小鼠CFS具有明显的治疗作用。
Objective: To observe the antagonistic effect and its related mechanism of water extract on chronic fatigue syndrome (CFS) induced by stress in mice. Methods: CFS model was established by forced cold water swimming stress. From stress point of view, we observed the general behavior, hypothalamic-pituitary-adrenal axis (HPA axis) function changes, central nervous system neurotransmitter Changes and skeletal muscle energy metabolism indicators, and further explore the cress extract mechanism of chronic fatigue. Results: Cold water stress significantly decreased spontaneous activity in mice; HPA axis function index: serum ACTH, CORT and sex hormone testosterone (T) decreased; central neurotransmitter index: 5 (5-HT) and dopamine (DA) increased significantly. The energy metabolism index of skeletal muscle in peripheral fatigue: the activity of ultramicrosine Ca2 + -Mg2 + -ATP decreased, while the content of phosphate (Pi) (MDA) increased significantly, superoxide dismutase activity (SOD) decreased significantly. After drug treatment, the autonomic ability of mice increased obviously, the function of HPA axis and gonadal axis returned to normal, the central nervous system neurotransmitter decreased, peripheral fatigue improved and the free radicals in the body were eliminated to the normal level. Conclusion: Cress extract has obvious therapeutic effect on mouse CFS.
