目的 :进一步研究SLE病人的ds DNA抗体重链V区基因的组成特点及其VH基因区突变在系统性红斑狼疮狼疮肾炎的发病机理中的作用。方法 :从SLE肾炎病人外周血中分离淋巴细胞制备的噬菌抗体库中克隆出ds DNA单克隆抗体细胞株NYE3,对其IgV基因进行检测和分析。结果 :NYE3单克隆抗体VH在编基因的CDR区出一精氨酸和赖氨酸等带正电荷的氨基酸突变。结论 :SLE病人的ds DNA抗体的VH编码基因的CDR区出现带正电荷的氨基酸突变改变可能与SLE患者肾炎的发病有关。
OBJECTIVE: To further study the composition of the heavy chain V region gene of ds DNA antibody and the role of VH gene mutation in the pathogenesis of systemic lupus erythematosus lupus nephritis. METHODS: The dsDNA monoclonal antibody cell strain NYE3 was cloned from the phage antibody library prepared from lymphocytes isolated from the peripheral blood of patients with SLE nephritis. The IgV gene was detected and analyzed. Results: The NYE3 monoclonal antibody VH mutated a positively charged amino acid such as arginine and lysine in the CDR regions of the coding genes. CONCLUSIONS: The positive amino acid mutations in the CDRs of the VH-encoding genes of ds DNA antibodies in patients with SLE may be related to the pathogenesis of nephritis in SLE patients.
