本文研究比较了三种β-内酰胺酶抑制剂:棒酸、CP-45899、UK-38006,对从不同菌株制备的各类β-内酰胺酶提取液及细胞内酶的的抑酶作用。实验所用10种β-内酰胺酶均经等电聚焦法鉴定。酶抑制剂的相对抑酶作用用紫外分光光度法与生物法测定。 研究结果表明棒酸与UK-38006在浓度低于1μg/ml时即能抑制大多数β-内酰胺酶,CP-45899抑酶作用不如前二种化合物。对Whatman酶,UK-38006作用最强,对KI酶,则以棒酸作用最明显。棒酸与羧苄青霉素或呋苄青霉素联合,对细胞内酶呈现最佳协同作用。
In this paper, three kinds of β-lactamase inhibitors: clavulanic acid, CP-45899, UK-38006 were compared and their inhibitory activities against various β-lactamase extracts and intracellular enzymes prepared from different strains were compared. The 10 β-lactamases used in the experiment were identified by isoelectric focusing. The relative inhibitory effect of enzyme inhibitor was determined by UV spectrophotometry and bioassay. The results showed that clavulanic acid and UK-38006 inhibited most β-lactamases at concentrations below 1 μg / ml. CP-45899 exhibited less inhibition than the first two compounds. For Whatman enzyme, UK-38006 the strongest role, the KI enzyme, clavulanic acid role is most obvious. Clavulanic acid combined with carbenicillin or furcillin exert the best synergistic effect on intracellular enzymes.
