目的研究子痫前期(preeclampsia,PE)患者胎盘组织中的黏蛋白1(MUC1)、整合素β3(Integrinβ3)的表达情况,探讨二者在子痫前期胎盘中的作用。方法采用蛋白质印迹法检测子痫前期轻度组(轻度PE组,20例)、重度组(重度PE组,20例)和正常妊娠组(正常组,20例)胎盘组织中MUC1和Integrinβ3蛋白的表达。结果 1 MUC1在正常组,轻度PE组和重度PE组胎盘组织中相对表达量分别为0.53±0.13、0.56±0.12和0.90±0.20,轻、重度PE组均明显高于正常组(P<0.05);2正常组、轻度和重度PE组胎盘组织中Integrinβ3的相对表达量分别为0.61±0.14、0.57±0.13和0.51±0.12,轻、重度PE组表达均低于正常组(P<0.05);3 MUC1与Integrinβ3在子痫前期患者胎盘中的表达呈负相关(r=-0.79,P<0.05)。结论 MUC1蛋白在子痫前期的表达随病情加重而增加,Integrinβ3蛋白的表达随病情的加重而降低,二者可能共同参与子痫前期发病。
Objective To study the expression of MUC1 and Integrinβ3 in the placenta of preeclampsia (PE) and to explore their roles in placenta preeclampsia. Methods Western blotting was used to detect the expression of MUC1 and Integrinβ3 in mild preeclampsia group (mild PE group, 20 cases), severe cases (severe PE group, 20 cases) and normal pregnancy group (20 cases) expression. Results 1 The relative expression levels of MUC1 in placental tissues of normal, mild PE and severe PE groups were 0.53 ± 0.13, 0.56 ± 0.12 and 0.90 ± 0.20, respectively. The levels of MUC1 in mild and severe PE group were significantly higher than those in normal group (P <0.05) ). 2 The relative expression of Integrinβ3 in normal and mild and severe PE groups were 0.61 ± 0.14,0.57 ± 0.13 and 0.51 ± 0.12, respectively. The expression of integrinβ3 in mild and severe PE group was lower than that in normal group (P <0.05) ; 3 There was a negative correlation between the expression of MUC1 and Integrinβ3 in the placenta of preeclampsia (r = -0.79, P <0.05). Conclusions The expression of MUC1 in preeclampsia increases with the progression of the disease. The expression of Integrinβ3 decreases with the progression of the disease, both of which may participate in the pathogenesis of preeclampsia.
