目的检测头颈鳞癌标本,寻找与头颈鳞癌转移高度相关的基因。方法采用单核苷酸多态性基因芯片技术,检测80例临床头颈鳞癌标本,包括舌癌17例,口腔癌13例,口咽癌14例,下咽癌36例。在随访期内,将发生远处转移的39例患者标本作为试验组,将未发生远处转移的41例患者标本作为对照组,利用Cox回归分析基因单核苷酸多态性对肿瘤远处转移的影响。结果单核苷酸多态性检测发现,在位于11q13的区域出现了一个基因组增多所致的独特的峰值。在这个基因组内,Cox回归分析显示,SHANK2基因的rs9651738位点的T等位基因相对G等位基因的相对危险度是3.61(P=0.003),FGF4基因的rs4980690位点的T等位基因相对G等位基因的相对危险度是3.63(P=0.006),ANO1基因的rs7929885位点的T等位基因相对G等位基因的相对危险度是4.35(P=0.008),PPFIA1基因的rs687660位点的T等位基因相对G等位基因的相对危险度是4.24(P=0.011),ORAOV1基因的rs6606651位点的T等位基因相对G等位基因的相对危险度是3.04(P=0.013),CCND1基因的rs592412位点的T等位基因相对G等位基因的相对危险度是4.07(P=0.013)。结论位于基因组11q13的SHANK2基因的rs9651738位点、FGF4基因的rs4980690位点、ANO1基因的rs7929885位点、PPFIA1基因的rs687660位点、ORAOV1基因的rs6606651位点、CCND1基因的rs592412位点的T等位基因相对G等位基因可增加头颈鳞癌发生转移的危险性。
Objective To detect specimens of squamous cell carcinoma of head and neck and to search for genes highly correlated with the metastasis of head and neck squamous cell carcinoma. Methods Eighty clinical samples of squamous cell carcinoma of head and neck were detected by single nucleotide polymorphism gene chip technique, including 17 cases of tongue cancer, 13 cases of oral cancer, 14 cases of oropharyngeal carcinoma and 36 cases of hypopharyngeal carcinoma.
