“干柱”层析是一种改进的层析技术。运用这种方法在一根柱上能很迅速地进行制备性分离,其分离效果可与薄层层析相比。该法可用于薄层层析能够分离的任何混合物,包括那些通常以“湿装”(liquid-filled)柱不能分离的物质。已知:(1)降活性的吸附剂比高活性吸附剂的分离效果好得多;(2)“干柱”层析的分离效果和薄层层析一样好。(3)“干柱”层柱的分离直接与薄层层析有关联(一个重要的关系是薄层层析的条件可以直接套用到“干柱”层析上);(4)制备性分离所需柱的大小可以预知。“干柱”层析已成功地用于分离甾体、生物碱、类脂质、酸类、胺类和多种杂环化合物的混合物,并已用于分离放射性物质和代谢
Dry column chromatography is an improved chromatography technique. Using this method, preparative separations can be performed very quickly on a column with comparable separation as thin-layer chromatography. This method can be used for any mixture that can be separated by thin layer chromatography, including those that can not be separated by a “liquid-filled” column. It is known that: (1) deactivating sorbents are much better than high activity sorbents; and (2) “dry column” chromatography is as good as thin layer chromatography. (3) The separation of “dry column” pillars is directly related to thin layer chromatography (an important relationship is that the conditions of thin layer chromatography can be applied directly to “dry column” chromatography); (4) preparative separation The size of the required columns can be predicted. “Dry column” chromatography has been successfully used to separate mixtures of steroids, alkaloids, lipids, acids, amines and various heterocyclic compounds and has been used to separate radioactive materials and metabolites
