对6只猕猴,每天2次(8:00、17:00)肌肉注射泰国4号海洛因水溶液(100g/L ),第1天剂量为0.5ml,每3天增加0.25ml,到第15天止。给药初期出现震颤、嗜睡、昏沉等体征。实验过程中每天称猴的体重,发现体重急骤下降。第6天和第14天注射前抽血5ml,检测吗啡、血糖和肝功能,发现猴血糖及ALT、AST均升高;提示海洛因对猴的糖代谢有影响,对肝功能有损害作用,在血液中分别测得一定量的吗啡,可作成瘾判定指标。第15天,用纳洛酮催引,结果出现明显的戒断症状,表明模型已成功建立。
