Objective:To observetheexpressionof humanleukocyticantigenDR(HLA-DR)inprimaryhepatocellular carcinoma(HCC)anditsup-regulationby interferon(IFN).Methods:The expressionof HLA-DRin46specimensof humanHCCtissues,4humanHCCcelllines(SMMC-7721,HCC-9204,BEL-7402andHHCC)anda humanhepatocyte celllineQZGwas respectivelydetectedby immunohistochemicalABCstainingandflowcytometry.Theexpressionof HLA-DRinthe5celllineswasdetectedby ELISAbeforeandafterthecellsweretreatedwithIFN-γor IFN-α.Results:Eighteenoutof46HCCtissues(39.1%)expressedHLA-DR,whereasallthenormallivertissuesimmediatelyadjacentto HCCtissueswereHLA-DR-negative.No obviousHLA-DR-positivestainingwas foundin allthe5celllines.The expressionof HLA-DRwas up-regulatedin allthe5celllinesafterIFN-γor IFN-αtreatment.The up-regulationof HLA-DRinQZGcellswaslessobviousthanthatinHCCcelllines.Theeffectof IFN-γwasmoresignificantthanthatof IFN-α.Conclusion:HCCtissuescanexpressHLA-DRto someextent,butHCCcelllinesdo notexpressdetectable HLA-DR.IFNcanup-regulateHLA-DRexpressioninHCCcells.
