目的:探讨环氧化酶-2(cyclooxygenase-2,COX-2)基因及蛋白在乳腺癌中的表达及临床意义。方法:采用原位杂交和S-P免疫组织化学技术检测20例乳腺纤维腺瘤和52例乳腺癌中COX-2mRNA和蛋白的表达。结果:COX-2mRNA和蛋白在乳腺纤维腺瘤和乳腺癌组织中的阳性率分别为(20.0%、65.4%)、(15.0%、57.7%),差异有显著性(P<0.01);COX-2mRNA和蛋白表达之间差异无显著性(P>0.05)。COX-2mRNA表达与淋巴结转移和肿瘤大小有关(P<0.05),与年龄和组织学分级无关(P>0.05);COX-2蛋白表达与淋巴结转移有关(P<0.05),与年龄、肿瘤大小和组织学分级无关(P>0.05)。结论:(1)COX-2mRNA及蛋白在乳腺癌中表达上调,提示COX-2在乳腺癌的发生、发展中起重要作用。(2)COX-2与淋巴结转移正相关,提示COX-2可作为判断乳腺癌预后的标记物。
Objective: To investigate the expression and clinical significance of cyclooxygenase-2 (COX-2) gene and protein in breast cancer. Methods: The expressions of COX-2 mRNA and protein in 20 cases of breast fibroadenoma and 52 cases of breast cancer were detected by in situ hybridization and S-P immunohistochemistry. RESULTS: The positive rates of COX-2 mRNA and protein in breast fibroadenoma and breast cancer were (20.0%, 65.4%), (15.0%, 57.7%) respectively, with significant difference (P <0.01) There was no significant difference between 2 mRNA and protein expression (P> 0.05). The expression of COX-2 mRNA was correlated with lymph node metastasis and tumor size (P <0.05), but not with age and histological grade (P> 0.05). COX-2 protein expression was correlated with lymph node metastasis And histological grade unrelated (P> 0.05). CONCLUSION: (1) COX-2 mRNA and protein are upregulated in breast cancer, suggesting that COX-2 plays an important role in the occurrence and development of breast cancer. (2) The positive correlation between COX-2 and lymph node metastasis suggested that COX-2 could be used as a marker to predict the prognosis of breast cancer.
