目的:通过对铜绿假单胞菌在溴化十六烷基三甲铵琼脂培养基中生长性能差异的分析,建立培养基的评价分析方法。方法:利用96孔板设置不同培养基处方(3种胨源×2种培养基配方)、溴化十六烷基三甲铵浓度(6水平)和铜绿假单胞菌浓度(4水平)的组合,分析在各种不同的组合条件下铜绿假单胞菌的生长情况。结果:不同溴化十六烷基三甲铵琼脂促生长能力的差异主要由胨源和配方所决定;USP/EP收载的溴化十六烷基三甲铵琼脂配方优于目前中国药典收载的配方。结论:多因素多水平的微量分析方法可较好地揭示影响培养基促生长能力的诸因素,为培养基配方的筛选开拓新思路。
OBJECTIVE: To establish a method for the evaluation of culture medium by analyzing the difference of growth performance of Pseudomonas aeruginosa in cetyltrimethylammonium bromide (BGC) agar medium. Methods: The combination of different media (three kinds of peptone source and two kinds of culture medium), Cetyltrimethylammonium bromide concentration (six levels) and Pseudomonas aeruginosa concentration (four levels) were set up in 96-well plates , Analysis of the growth of Pseudomonas aeruginosa under various combinations of conditions. Results: The differences in the growth promoting ability of cetyltrimethylammonium bromide were mainly determined by the peptone source and the formula. The formula of cetyltrimethylammonium bromide (ACP) loaded by USP / EP was better than that of the Chinese Pharmacopoeia formula. CONCLUSION: The multi-factor and multi-level microanalysis method can better reveal the factors that affect the growth-promoting ability of the medium and open up new ideas for the screening of the medium formula.
