目的验证蛋白质组学筛选出的慢性砷暴露血清差异表达蛋白——玻璃粘连蛋白(vitronectin)含量,为研究砷暴露人群血管疾病的发生提供线索。方法采用酶联免疫吸附试验(ELISA),验证筛选出的vitronectin在不同砷暴露剂量组的表达。结果对照组(228.45±34.14 ng/ml)和低(219.31±28.73 ng/ml)、高(199.84±29.11 ng/ml)剂量砷暴露组vitronectin血清含量差异有统计学意义(F=128.00,P<0.01),水砷含量与血清vitronectin水平呈负相关(OR=-383.95)。结论初步验证vitronectin可作为砷暴露致健康损害的血清生物标志物。
Objective To verify the expression of vitronectin, a differentially expressed protein of chronic arsenic exposed by proteomics, to provide clues for studying the occurrence of vascular diseases in arsenic-exposed population. Methods The enzyme-linked immunosorbent assay (ELISA) was used to verify the expression of vitronectin in different doses of arsenic exposure group. Results The vitronectin levels in the arsenic exposure group (228.45 ± 34.14 ng / ml) and the low (219.31 ± 28.73 ng / ml) and high (199.84 ± 29.11 ng / ml) groups were significantly different (F = 128.00, P < 0.01). The content of arsenic in water was negatively correlated with the level of vitronectin (OR = -383.95). Conclusion Preliminary validation of vitronectin as a biomarker of health damage caused by arsenic exposure.
