目的通过研究共轭亚油酸(conjugated linoleic acid,CLA)对饮食诱导肥胖大鼠脂肪酸连接蛋白(fatty acid binding proteins,ap2)基因表达的影响,探讨CLA抗糖尿病作用的机制。方法选用雄性Wistar大鼠,随机分为对照组、高脂组、高脂+CLA组(每100g饲料含CLA分别为0.75g、1.50g、3.00g),每组动物10只,观察CLA对肥胖大鼠胰岛素、血糖水平的影响,并应用RT-PCR的方法检测ap2、过氧化物酶体增殖物激活受体γ(PPARγ)的表达水平。结果高脂组大鼠血清游离脂肪酸(free fatty acids,FFA)、胰岛素和糖血水平分别为0.74±0.05(mmol/L)、11.11±2.73 (μIU/ml),5.09±0.66(mmol/L),CLA可降低肥胖大鼠血清FFA、血糖、胰岛素水平,低、中、高剂量组FFA水平分别为0.69±0.05(mmol/L)、0.61±0.06(mmol/L)、0.58±0.06(mmol/L),胰岛素水平分别为6.99±1.77(μIU/ml),7.36±1.48(μIU/ml),7.85±1.60(μIU/ml)(P<0.05),血糖水平分别为4.28±0.72(mmol/L),4.18±0.55(mmol/L)(P<0.05),4.06±0.63(mmol/L)(P<0.05),且CLA可增加肥胖大鼠脂肪组织ap2、PPARγmRNA的表达水平。结论CLA可通过激活PPARγ上调脂肪酸连接蛋白基因的表达,改善肥胖大鼠的胰岛素抵抗。
Objective To investigate the effect of conjugated linoleic acid (CLA) on the expression of fatty acid binding proteins (ap2) in diet-induced obese rats and to explore the mechanism of anti-diabetic effects of CLA. Methods Male Wistar rats were randomly divided into control group, high fat group, high fat + CLA group (CLA, 0.75g, 1.50g, 3.00g per 100g diet), 10 animals in each group. Rats, insulin and blood glucose levels. The expression levels of ap2 and peroxisome proliferator-activated receptor γ (PPARγ) were detected by RT-PCR. Results The serum levels of free fatty acids (FFA), insulin and glycemia in high fat diet group were 0.74 ± 0.05 (mmol / L), 11.11 ± 2.73 (μIU / ml) and 5.09 ± 0.66 (mmol / L) , CLA reduced serum FFA, blood glucose and insulin levels in obese rats. The levels of FFA in low, middle and high dose groups were 0.69 ± 0.05 (mmol / L), 0.61 ± 0.06 (mmol / L) and 0.58 ± 0.06 (P <0.05). The blood glucose levels were respectively 4.28 ± 0.72 (mmol / L) and insulin levels were 6.99 ± 1.77 (μIU / ml), 7.36 ± 1.48 (μIU / ml) and 7.85 ± 1.60 , 4.18 ± 0.55 (mmol / L), 4.06 ± 0.63 (mmol / L), respectively (P <0.05), and CLA could increase the expression of ap2 and PPARγmRNA in adipose tissue of obese rats. Conclusion CLA can improve insulin resistance in obese rats by activating the expression of fatty acid connexin gene by PPARγ.
