目的研究苯并咪唑对体外培养大鼠睾丸支持细胞波形蛋白和微管蛋白表达的影响。方法取18~20日龄Wistar雄性大鼠睾丸支持细胞,通过Tris-Hcl低渗处理进行细胞纯化,用MTT法检测阴性对照组、溶剂对照组、苯并咪唑剂量组(0.1、1、10和100μg/ml)中大鼠睾丸支持细胞的生长抑制率。用免疫细胞化学法检测细胞中波形蛋白、α/β-微管蛋白的表达。结果 10和100μg/ml苯并咪唑剂量组在24、48和72 h时均明显抑制了支持细胞的增殖;10和100μg/ml苯并咪唑剂量组中支持细胞波形蛋白、α/β-微管蛋白表达明显减少。结论苯并咪唑对体外培养大鼠支持细胞毒作用明显,抑制睾丸支持细胞的增殖,影响波形蛋白、α/β-微管蛋白的表达。
Objective To investigate the effect of benzimidazole on the expression of vimentin and tubulin in cultured rat testicular sertoli cells. Methods Sertoli cells from 18 to 20-day-old Wistar male rats were isolated and purified by tris-Hcl hypotonic treatment. MTT assay was used to detect the inhibitory effect of negative control group, solvent control group, benzimidazole dose group (0.1, 100μg / ml) in rat testicular support cell growth inhibition rate. Immunocytochemistry was used to detect the expression of vimentin and α / β-tubulin in the cells. Results Both benzalkonium chloride group and 10, 100μg / ml benzimidazole group significantly inhibited the proliferation of support cells at 24h, 48h and 72h. In the 10 and 100μg / ml benzimidazole dose groups, Protein expression was significantly reduced. Conclusions Benzimidazole can significantly inhibit the proliferation of rat supporting cells and inhibit the expression of vimentin and α / β-tubulin.
