目的:研究急性给予甲基苯丙胺(MAP)对脂多糖(LPS)诱导的小鼠机体产生细胞因子IL-6,IL-10的影响以及多巴胺(DA)D3受体可能参与的调节作用。方法:野生型(D3+/+)小鼠20只和DAD3受体敲除型(D3-/-)小鼠18只,体重20g±s2g,随机分组,♂♀各半,先腹腔注射(ip)NS(生理盐水,0.01ml.g-1)或MAP(5mg·kg-1即0.01ml.g-1),间隔10min后,ipLPS(150μg·kg-1即0.01ml.g-1)刺激,间隔1h后,眼球采血,血样4℃过夜,离心分离血清,用R&D公司ELISA试剂盒检测IL-6,IL-10的含量。结果:野生型(D3+/+)小鼠,MAP+LPS组(n=10)与NS+LPS组(n=10)相比较,血清IL-6含量几乎没有变化,但是血清中IL-10含量显著升高,并且差异有显著统计学意义(P<0.01)。DAD3受体敲除型(D3-/-)小鼠,MAP+LPS组(n=9)与NS+LPS组(n=9)相比较,血清IL-6含量下降,但差异无统计学意义(P=0.13)。DAD3受体敲除型(D3-/-)小鼠,NS+LPS组(n=9)与野生型(D3+/+)小鼠,NS+LPS组(n=10)相比较,血清IL-6含量轻微下降,差异无统计学意义(P=0.29)。DAD3受体敲除型(D3-/-)小鼠,MAP+LPS组(n=9)与野生型(D3+/+)小鼠MAP+LPS组(n=10)相比较,血清IL-6含量显著下降,并且差异有显著统计学意义(P<0.01)。结论:(1)MAP急性预处理后,小鼠血清IL-10含量显著升高,MAP可能是通过促进IL-10的产生从而发生免疫功能抑制的。(2)MAP急性预处理后,DAD3受体敲除型(D3-/-)小鼠MAP+LPS组与野生型(D3+/+)小鼠MAP+LPS组相比较,血清IL-6含量显著下降,DAD3受体对LPS诱导的小鼠产生IL-6可能有正向调控作用。
Objective: To investigate the effects of acute methamphetamine (MAP) on IL-6 and IL-10 induced by lipopolysaccharide (LPS) in mice and the possible involvement of dopamine D3 receptors. METHODS: Twenty (20) wild type (D3 + / +) mice and 18 DAD3 receptor knockout (D3 - / -) mice weighing 20 g ± s2 g were randomly divided into three groups: NS (normal saline, 0.01ml.g-1) or MAP (5mg · kg-1, 0.01ml.g-1). After 10min interval, ipLPS (150μg · kg- After an interval of 1h, eyeball blood sampling, blood samples overnight at 4 ℃, centrifuged serum, R & D ELISA kit detection of IL-6, IL-10 content. Results: Compared with NS + LPS group (n = 10), there was almost no change in serum IL-6 level in wild type (D3 + / +) mice and MAP + LPS group Significantly increased, and the difference was statistically significant (P <0.01). Compared with NS + LPS group (n = 9), the level of IL-6 in DAD3 receptor knock-out (D3 - / -) mice and MAP + LPS group (n = 9) decreased but the difference was not statistically significant (P = 0.13). Compared with D3 + / + mice and NS + LPS group (n = 10), the levels of IL-6β in the DAD3 receptor knockout (D3 - / - 6 content decreased slightly, the difference was not statistically significant (P = 0.29). Compared with the MAP + LPS group (n = 10) in the wild type (D3 + / +) mice, the levels of IL-6 in the DAD3 receptor knockout (D3 - / - Content decreased significantly, and the difference was statistically significant (P <0.01). Conclusions: (1) After acute MAP preconditioning, the level of IL-10 in serum of mice is significantly increased. MAP may inhibit the immune function by promoting the production of IL-10. (2) Compared with the MAP + LPS group of wild-type (D3 + / +) mice, MAP-LPS group of DAD3 receptor knockout (D3 - / - DAD3 receptor may have a positive regulatory effect on LPS-induced IL-6 production in mice.
