目的:探讨不同浓度白藜芦醇在常氧环境及缺氧环境下对肝癌细胞株BEL-7402增殖及凋亡的影响。方法:采用MTT法分析不同浓度(0、12.5、25、50、100、200μmol/L)白藜芦醇在常氧环境及缺氧环境下对肝癌细胞株BEL-7402增殖的影响,应用流式细胞仪检测其对BEL-7402细胞凋亡的影响。结果:不同浓度的白藜芦醇在常氧及缺氧环境中对肝癌细胞株BEL-7402均有明显抑制增殖及诱导凋亡作用,呈剂量-效应相关,其中50、100、200μmol/L白藜芦醇两种环境中对肝癌细胞的抑制率及凋亡率与对照组比较差异均具有统计学意义(P<0.05)。50、100、200μmol/L白藜芦醇,常氧环境中对肝癌细胞的抑制率分别为18.70±2.15%、22.13±2.18%、31.47±2.26%,凋亡率分别为11.82±2.28%、19.80±2.64%、38.00±2.57%;缺氧环境中对肝癌细胞的抑制率分别为17.87±1.76%、23.52±2.29%、29.87±1.51%,凋亡率分别为12.36±3.15%、19.71±2.78%、37.87±4.83%。结论:白藜芦醇在缺氧环境下能抑制肝癌细胞株BEL-7402的增殖并诱导其凋亡,且其作用与常氧环境相似。
Objective: To investigate the effects of different concentrations of resveratrol on proliferation and apoptosis of hepatocellular carcinoma cell line BEL-7402 under normoxic and anoxic conditions. Methods: The effects of different concentrations of resveratrol (0,12.5,25,50,100,200 μmol / L) on the proliferation of hepatocellular carcinoma cell line BEL-7402 under normoxic and anoxic conditions were analyzed by MTT assay. The effect of BEL-7402 on the apoptosis of BEL-7402 cells was detected by cytometry. RESULTS: Different concentrations of resveratrol significantly inhibited the proliferation and induced apoptosis of hepatocellular carcinoma cell line BEL-7402 under normoxia and hypoxia conditions in a dose-dependent manner. Among them, 50,100 and 200μmol / L of white The inhibitory rate and apoptosis rate of hepatoma cells in two resveratrol groups were significantly different from those in control group (P <0.05). 50, 100, 200μmol / L resveratrol, respectively. CONCLUSION: Resveratrol inhibits the proliferation and induces apoptosis of hepatocellular carcinoma cell line BEL-7402 in anoxic environment, and its effect is similar to that of normoxic environment.
