目的:研究脑缺血中氧自由基(OFR)和一氧化氮(NO)的动态变化及金丝桃甙(Hyp)的作用。方法:脑缺血模型由结扎大鼠四血管造成。用低温电子顺磁共振(ESR)技术和gren法分别测定脑皮质中的OFR和NO。结果:脑缺血5min时,NO就增多,15min时,OFR才有增多;50,100mg·kg-1Hyp显著减少,OFR和NO的增高,抑制超氧化物歧化酶和乳酸脱氧酶活性的下降,延长小鼠断头后张口喘气时间。结论:脑缺血中,NO增多早于OFR,Hyp对脑缺血损伤有保护作用。
Objective: To study the dynamic changes of oxygen free radical (OFR) and nitric oxide (NO) during cerebral ischemia and the effect of hypericum perforatum (Hyp). METHODS: A model of cerebral ischemia was caused by ligation of four blood vessels in rats. The OFR and NO in cerebral cortex were measured by low-temperature electron paramagnetic resonance (ESR) technique and gren method. RESULTS: At 5 min after cerebral ischemia, NO increased. At 15 min, the OFR increased; 50, 100 mg·kg-1 Hyp decreased significantly, OFR and NO increased, and the activities of superoxide dismutase and lactate deoxygenase were inhibited. Extend the mouth breathing time after the mice become decapitated. Conclusion: In cerebral ischemia, the increase of NO is earlier than that of OFR. Hyp has protective effect on cerebral ischemic injury.
