目的了解大鼠高压电烧伤后血清CD44变化,探讨乌司他丁(UTI)对其的干预效果。方法将180只清洁级大鼠按随机数字表法分为对照组、电伤组和治疗组,每组60只,各组每时相点10只。电伤组和治疗组大鼠用调压器及实验变压器制成高压电烧伤模型,对照组接相同装置但不通电致假伤。于伤前15 min及伤后5 min、1 h、2 h、4 h、8 h,大鼠开胸后在直视下心脏采血,分离血清,用ELISA双抗体夹心法检测血清CD44含量。对数据行两因素析因设计的方差分析和LSD检验。结果①对照组和电伤组大鼠血清CD44含量的比较:电伤组CD44含量高于对照组(主效应F=882.901,P<0.01);电伤组CD44含量受伤后时间变量影响(主效应F=81.459,P<0.01),伤后5 min~8 h各时相均高于本组伤前值(P<0.01),且呈逐渐增高趋势。②电伤组和治疗组大鼠血清CD44含量的比较:治疗组CD44含量低于电伤组(主效应F=219.597,P<0.01);治疗组CD44含量受伤后时间变量影响(主效应F=155.774,P<0.01),治疗组伤后5 min~8 h各时相均高于本组伤前值(P<0.01)。结论高压电烧伤引起大鼠血清CD44含量升高,而UTI能降低高压电烧伤后的大鼠血清CD44含量。
Objective To investigate the changes of serum CD44 after high voltage electric burn in rats and investigate the effect of UTI on it. Methods 180 clean-grade rats were randomly divided into control group, electric injury group and treatment group according to random number table method, 60 rats in each group, with 10 rats in each group. The electric injury group and the treatment group rats were made into high-voltage electric burn model by the voltage regulator and experimental transformer, and the control group were connected to the same device but were not electrified. Serum CD44 levels were measured by ELISA double antibody sandwich method after 15 min and 5 min, 1 h, 2 h, 4 h and 8 h after injury. Analysis of variance and LSD test on two factors factorial design of data line. Results ① Compared with the control group, the content of CD44 in the control group and the electric injury group were significantly higher than those in the control group (main effect F = 882.901, P <0.01) F = 81.459, P <0.01). The time from 5 min to 8 h after injury was higher than the pre-injury value (P <0.01), and showed an increasing trend. ② The level of serum CD44 in electric injury group and treatment group was lower than that in electric injury group (main effect F = 219.597, P <0.01); The time variable of the content of CD44 in the treatment group was influenced by time variable (main effect F = 155.774, P <0.01). The time of treatment group from 5 min to 8 h after injury was higher than that before injury (P <0.01). Conclusion High-voltage electric burn can increase the level of serum CD44 in rats, while UTI can reduce the level of serum CD44 in rats after high-voltage electric burn.
