探讨四物汤预防和治疗血虚证的作用机理。用6 0 Coγ射线全身照射小鼠造成血虚证模型 ;荧光标记并用流式细胞仪测定CD34+ 细胞在骨髓有核细胞中的比例 ;细胞固定后PI染色测定细胞周期 ;AnnexinV FITC试剂盒和DNA凝胶电泳检测骨髓细胞的凋亡。结果 :四物汤对小鼠骨髓造血干祖细胞、外周血细胞的恢复有明显促进作用 ;有保护骨髓和促进骨髓细胞由G1期进入S期的效应 ;四物汤可减轻射线引起的细胞凋亡 ;促进骨髓造血干祖细胞的增殖 ,抑制骨髓造血干祖细胞的凋亡是四物汤预防和治疗血虚证的机制之一
To explore the mechanism of Siwu Decoction on prevention and treatment of blood deficiency syndrome. Whole blood irradiation of mice with 60 Co γ-rays resulted in a blood deficiency model; the proportion of CD34+ cells in bone marrow nucleated cells was measured by fluorescence labeling and flow cytometry; cell cycle was determined by PI staining after cell fixation; AnnexinV FITC kit and DNA clotting Gel Electrophoresis Detects Apoptosis of Bone Marrow Cells RESULTS: Siwu decoction promoted the recovery of bone marrow hematopoietic stem and progenitor cells and peripheral blood cells in mice; it had the effect of protecting bone marrow and promoting the progression of bone marrow cells from G1 phase to S phase; Siwu decoction could reduce radiation-induced apoptosis. Promoting the proliferation of bone marrow hematopoietic stem and progenitor cells and inhibiting the apoptosis of bone marrow hematopoietic stem and progenitor cells is one of the mechanisms of Siwu decoction in the prevention and treatment of blood deficiency syndrome
