目的:研究三氧化二砷(As2 O3 )注射液对人结肠癌裸鼠移植瘤端粒酶活性及其催化亚单位人端粒逆转录酶(hTERT)表达的影响。方法:建立人结肠癌裸鼠移植瘤模型,随机分为生理盐水组、As2 O3 低剂量(2 .5mg·kg-1)组、As2 O3 高剂量(5mg·kg-1)组及5 - FU(2 0mg·kg-1)组。腹腔注射As2 O3 ,利用银染 端粒重复扩增(TRAP)法测定人结肠癌裸鼠移植瘤端粒酶活性、RT PCR法检测hTERT的表达。结果:生理盐水组及5 -FU组端粒酶活性及其催化亚单位表达不受影响,不同剂量As2 O3 均明显抑制端粒酶活性及其催化亚单位表达。结论:As2 O3 注射液对人结肠癌裸鼠移植瘤端粒酶活性及其催化亚单位的表达有抑制作用。
Objective: To study the effect of As2O3 on the telomerase activity and the expression of catalytic subunit human telomerase reverse transcriptase (hTERT) in human colon carcinoma xenografts. Methods: The human colon cancer xenografts in nude mice were established and randomly divided into normal saline group, low dose As2 O3 (2.5 mg · kg -1) group, high dose As 2 O 3 (5 mg · kg -1) (20mg · kg-1) group. As2 O3 was injected intraperitoneally. The telomerase activity of human colon cancer xenografts in nude mice was detected by silver staining of telomeric repeat amplification (TRAP) method. The expression of hTERT was detected by RT PCR. Results: The telomerase activity and the expression of its catalytic subunit in the normal saline group and the 5-FU group were not affected. As2 O3 at different doses significantly inhibited telomerase activity and its catalytic subunit expression. Conclusion: As2 O3 injection can inhibit telomerase activity and the expression of catalytic subunit in human colon cancer xenografts.
