目的研究高碘摄入对仔鼠脑髓鞘化的影响及硒干预的作用。方法将Balb/c小鼠按体重随机分为3组:正常对照组、高碘组(3mg/L)以及高碘+Se组(3mg/L+0.5mg/L Se),分别喂饲高碘水或高碘-硒水以及正常鼠饲料。各组亲代鼠饲养4个月后雌雄按2:1交配,取14d仔鼠血、脑组织测定相关指标。结果与正常对照组相比,高碘仔鼠血清和脑组织TT_4显著下降,TT_3水平亦降低;脑MBP mRNA表达水平较对照组下降了27%。补硒提高仔鼠甲状腺激素的水平,上调仔鼠脑MBP mRNA的表达。结论高碘引起仔鼠甲状腺激素水平降低可能是仔鼠脑MBP mRNA表达水平下降的原因,硒可通过调节甲状腺激素水平从而上调仔鼠脑MBP mRNA的表达。
Objective To investigate the effect of high iodine intake on myelination in neonatal rats and the effect of selenium intervention. Methods Balb / c mice were randomly divided into three groups according to body weight: normal control group, high iodine group (3mg / L) and high iodine + Se group (3mg / L + 0.5mg / L Se) Water or iodine - selenium water and normal rat feed. After 4 months of feeding, the male and female of each group were mated by 2: 1, and the blood and brain tissue of 14 d were taken to measure the related indexes. Results Compared with the normal control group, the levels of TT_4 in serum and brain tissues were significantly decreased and the levels of TT_3 in hippocampus were also decreased. The expression of MBP mRNA in brain was decreased by 27% compared with the control group. Selenium supplementation increased thyroid hormone levels in offspring and up-regulated the expression of MBP mRNA in offspring. Conclusion The decrease of thyroid hormone in offspring induced by high iodine may be the reason of the decrease of MBP mRNA expression in offspring brain. Selenium can upregulate the expression of MBP mRNA in offspring brain by regulating thyroid hormone level.
