目的探讨漆黄素(fisetin)在体外对小鼠T淋巴细胞增殖、活化、凋亡、ROS的释放和巨噬细胞NO释放功能的影响。方法 CFDA-SE标记技术结合流式细胞术检测漆黄素对T淋巴细胞增殖及增殖指数(PI)的影响。荧光抗体染色结合流式细胞术分析漆黄素对T淋巴细胞在Con A的刺激下CD25的表达水平的影响。DIOC染色检测细胞线粒体膜电势的变化情况,H2DCFDA染色检测细胞ROS释放的变化。Griess试剂盒用以检测巨噬细胞NO释放功能。结果漆黄素(2.5,5和10μmol/L)明显抑制T淋巴细胞增殖,增殖指数(PI)降低并具有剂量依赖性。漆黄素可明显抑制CD25的表达(P<0.01)及ROS的释放。DiOC6(3)染色分析显示,漆黄素可促进细胞的凋亡。巨噬细胞在LPS和IFN-γ的刺激下的NO释放量明显增加,而在漆黄素作用下NO释放量明显下降(P<0.01)。结论漆黄素是一种潜在的免疫调节剂。
Objective To investigate the effects of fisetin on proliferation, activation, apoptosis, ROS release and NO release of macrophages in vitro. Methods The effect of enkephalin on the proliferation and proliferation index (PI) of T lymphocytes was detected by CFDA-SE and flow cytometry. Fluorescent antibody staining combined with flow cytometry was used to analyze the effect of enkephalin on the expression of CD25 on T lymphocytes stimulated by Con A. The changes of mitochondrial membrane potential were detected by DIOC staining. The change of ROS release was detected by H2DCFDA staining. Griess kit used to detect macrophage NO release function. Results Saponin (2.5, 5 and 10 μmol / L) significantly inhibited the proliferation of T lymphocytes, and the proliferation index (PI) decreased in a dose-dependent manner. Emodin significantly inhibited the expression of CD25 (P <0.01) and the release of ROS. DiOC6 (3) staining analysis showed that the emodin can promote cell apoptosis. The release of NO from macrophages stimulated by LPS and IFN-γ significantly increased, while the release of NO decreased markedly under the action of scutellarein (P <0.01). Conclusions Epigenin is a potential immunomodulator.
