雄性Sprague-Dawley(SD)大鼠,链脲佐菌素(STZ)造模,糖尿病(DM)成模后第12、16、20、24周,选取DM组分别为7、6、6、9只和NDM组各5只,测定晨尿白蛋白/肌酐比值及肾脏肥大指数,光学显微镜HE染色下观察肾小球及肾小管改变程度半定量评分,电子显微镜下测定GBM厚度。结果尿白蛋白/肌酐比值及肾脏肥大指数在DM组均明显高于NDM组,P<0.01,其中肾脏肥大指数DM组间20周高于12周及16周,P<0.05;24周高于12及16周,P<0.05及P<0.01。光镜下肾脏病理改变评分与GBM厚度均明显高于NDM组,P<0.01,其中光镜下肾脏病理改变评分DM组间20及24周升高明显;GBM厚度DM组间12、20、24周DM组高于NDM组,尤其24周明显高于16及20周,P<0.01。尿白蛋白/肌酐比值与GBM厚度在总的DM组及24周DM组均成正相关,P<0.05;肾脏肥大指数与光镜下肾脏病理改变评分在总的DM组正相关,P<0.05。结论尿白蛋白/肌酐比值、肾脏肥大指数、光镜下肾脏病理改变评分及GBM厚度在糖尿病SD大鼠中升高,反应肾脏损伤,其中24周病程变化更显著,尿白蛋白/肌酐比值与GBM厚度正相关,肾脏肥大指数与光镜下肾脏病理改变评分正相关。
Male Sprague-Dawley (SD) rats were given streptozotocin (STZ) and diabetes mellitus (DM) at the 12th, Only 5 rats in NDM group were given morning urine albumin / creatinine ratio and renal hypertrophy index. The degree of glomerular and tubular changes were observed under optical microscope. The thickness of GBM was determined by electron microscope. Results The urinary albumin / creatinine ratio and renal hypertrophy index in DM group were significantly higher than those in NDM group (P <0.01). The renal hypertrophy index in DM group was higher than that in DM group at 12 weeks and 16 weeks (P <0.05) 12 and 16 weeks, P <0.05 and P <0.01. The score of pathological changes of kidney and the thickness of GBM in light microscope were significantly higher than those in NDM group (P <0.01), and the score of pathological changes of kidney in DM group increased significantly at 20 and 24 weeks Week DM group was higher than NDM group, especially 24 weeks was significantly higher than 16 and 20 weeks, P <0.01. The urinary albumin / creatinine ratio was positively correlated with the thickness of GBM in total DM group and 24-week DM group, P <0.05. The renal hypertrophy index was positively correlated with the pathological change score of renal pathology in DM group (P <0.05). Conclusions The urinary albumin / creatinine ratio, renal hypertrophy index, pathological changes of kidney under light microscope and GBM thickness were increased in diabetic SD rats, which may lead to kidney damage. The course of the disease changes more significantly at 24 weeks, and the ratio of urinary albumin / creatinine GBM thickness was positively correlated, and the renal hypertrophy index was positively correlated with the pathological changes of kidney under light microscope.
