目的研究睫状神经营养因子(CNTF)对体外培养的大鼠大脑皮质星形胶质细胞(Ast)细胞周期及Fos蛋白表达的影响。方法通过体外纯化培养获取大鼠大脑皮质Ast,将CNTF加入培养液,作用相应的时刻点后,应用流式细胞仪测定细胞周期的变化,采用免疫细胞化学方法研究Fos蛋白的表达。结果CNTF作用Ast6 h后,可显著促进细胞周期进程,表现为G0/G1期细胞百分比降低;S期+G2/M期细胞百分比(增殖指数,poliferation index,PI值)升高。12h促增殖作用达高峰,24 h、48 h有所恢复,但PI仍明显高于对照组。CNTF作用于Ast 2 h后即可引起Fos蛋白的显著表达,并持续至24 h,而糖皮质激素预处理可明显抑制Fos蛋白表达。结论CNTF可促进Ast增殖及Fos蛋白表达。
Objective To investigate the effects of ciliary neurotrophic factor (CNTF) on cell cycle and Fos protein expression in cultured rat cortical astrocytes (Ast). Methods The rat cerebral cortex Ast was obtained by in vitro purification. The CNTF was added into the culture medium. After corresponding time points, the cell cycle was determined by flow cytometry. The expression of Fos protein was detected by immunocytochemistry. Results After 6 h of Astragalus, CNTF could significantly promote the cell cycle progression. The percentage of cells in G0 / G1 phase decreased and the percentage of cells in S phase and G2 / M phase increased (PI). 12h to promote proliferation reached a peak, 24 h, 48 h recovery, but PI was still significantly higher than the control group. CNTF can induce Fos protein expression in asthmatic rats for 2 h and continue to 24 h, while glucocorticoid pretreatment can significantly inhibit Fos protein expression. Conclusion CNTF can promote the proliferation of Ast and Fos protein expression.
