[目的]建立食品中蛋白质含量测定的离子色谱法。[方法]硫酸—双氧水体系消解样品,20mmoL/L的甲烷磺酸溶液作淋洗液,Ionpac CS12A色谱柱,DS-6电导检测器测定样品中蛋白质的含量。[结果]蛋白质(以NH4+-N计)在0.5~8.0mg/L范围内线性良好,方法检出限为0.21g/kg,样品加标回收率为93.1%~103%,相对标准偏差(RSD)为1.1%~3.2%。[结论]本方法简便、快速、准确,灵敏度及精密度高,用于食品中蛋白质含量的测定,结果满意。
[Objective] To establish a method for determination of protein in food by ion chromatography. [Method] The content of protein in samples was determined by digestion with sulfuric acid - hydrogen peroxide system, using 20 mmoL / L methanesulfonic acid solution as eluent, Ionpac CS12A column and DS - 6 conductivity detector. [Result] The results showed that the linear range was 0.5-8.0 mg / L for NH4 + -N, the detection limit was 0.21 g / kg and the spiked recoveries were 93.1% -103%. The relative standard deviation (RSD) ) Is 1.1% ~ 3.2%. [Conclusion] The method was simple, rapid, accurate, sensitive and precise, and was applied to the determination of protein in food with satisfactory results.
